Soybean is the most important source of vegetable oil and protein for human and animal diet in the world. The frequency of occurrence of drought has increased significantly over the last decade, probably associated with climate change occurred due to global warming. Researchers are looking
for a long time to improve and optimize the characteristics of the culture. Several strategies can be used to reduce losses caused by drought, ranging from adequate soil management and agriculture, to the use of irrigation. Another possibility is the development of cultivars adapted to drought
conditions and in this respect, Biotechnology has become an important tool in the development of varieties adapted to different conditions of biotic and abiotic stress. The genetically modified plants can be transformed by direct gene transfer using particle bombardment (biolistics), which inserts a
high number of copies of the transgene into the host genome, resulting many times in deactivation of the transgene by gene silencing. Moreover, the transformed plants via Agrobacterium tumefaciens generally have a low number of copies of the transgene. The use of this method allowed a stable expression and insertion of the interest gene in transgenic plants. The AREB is an important transcription factor signaled by the ABA, which acts in response to regulate the expression of other genes that are involved in tolerance to drought and high salinity stress. This study aimed to analyze the T1 plants transformed Agrobacterium by Southern blotting technique to detect the number of copies of the gene 35S:AREB1. The experiment was conducted at the Laboratory of Plant Biotechnology at Embrapa Soja, Londrina - PR. The process was performed according Kanamori et al., 2011. The Southern blotting was performed on 22 plants T1, AREB positive from two independent events. Of the 22 samples, 20 T1 plants had only one copy number and two plants had two copy numbers. Considering the complexity of the mechanism response to water deficit caused by drought, which can vary depending on the intensity and duration of stress, analysis on the molecular level, physiological and agronomic are needed later to assess the efficiency of the presence of the gene in transformed plants.
